Dojindo Molecular Technologies Inc DAPRed – Autophagy Detection (5 nmol)
DAPRed is used to detect autophagy in live cells. Autophagy is an intracellular degradation system, where dysfunctional proteins and organelles are degraded. In this process, aggregated dysfunctional proteins are surrounded by the double membrane to form an autophagosome. The small fluorescent molecule DAPRed is used to detect autophagosomes and autolysosomes. The mechanism suggests that the dye is incorporated into the autophagosome during double-membrane formation via structural features and then emits fluorescence under hydrophobic conditions. DALRed is cell-permeable, has no requirement of transfection method, and enables live-cell imaging with fluorescence microscopy. Excitation and emission of DAPRed: λex: 530 nm, λem: 720 nm. For monitoring only autolysosomes, we recommend DALGreen [D675] as it enables the detection of phagosome-lysosome fusion. Dojindo offers various reagents for autophagy research. Cell Biolabs’ OxiSelect™ Intracellular ROS Assay Kit is a cell-based assay for measuring hydroxyl, peroxyl, or other reactive oxygen species activity within a cell. The assay employs the cell-permeable fluorogenic probe 2’, 7’-Dichlorodihydrofluorescin diacetate (DCFH-DA). In brief, DCFH-DA is diffused into cells and is deacetylated by cellular esterases to non-fluorescent 2’, 7’-Dichlorodihydrofluorescin (DCFH), which is rapidly oxidized to highly fluorescent 2’, 7’-Dichlorodihydrofluorescein (DCF) by ROS. The fluorescence intensity is proportional to the ROS levels within the cell cytosol. The effect of antioxidant or free radical compounds on DCF-DA can be measured against the fluorescence of the provided DCF standard. The kit has a DCF detection sensitivity limit of 10 pM. Each kit provides sufficient reagents to perform up to 96 assays, including standard curve and unknown samples.
ELABSCIENCE Total Cholesterol (TC) Colorimetric Assay Kit (Single Reagent, COD-PAP Method) (E-BC-K109-M) 48 Tests
Total cholesterol includes free cholesterol and cholesterol esters. Cholesterol ester can be hydrolyzed by cholesterol esterase to produce cholesterol and free fatty acid. Cholesterol is oxidized by cholesterol oxidase to produce hydrogen peroxide. In the presence of Trinder's reagent, hydrogen peroxide is catalyzed by peroxidase (POD) to produce a red quinoneimine dye, whose color intensity is directly proportional to the TC concentration and can be measured at 510 nm.
